La m\u00e9thode utilise une tr\u00e8s grosse mol\u00e9cule appel\u00e9e EDTA qui forme un complexe avec les ions calcium. EDTA signifie acide \u00e9thyl\u00e8nediaminet\u00e9traac\u00e9tique. Un colorant bleu appel\u00e9 indicateur de Patton et Reeder (PR) est utilis\u00e9 comme indicateur. Ce colorant bleu forme \u00e9galement un complexe avec les ions calcium changeant de couleur du bleu au rose \/ rouge au cours du processus, mais le complexe colorant-ion m\u00e9tallique est moins stable que le complexe EDTA-ion m\u00e9tallique.
